Overview

In this study, six strains of recombinant African swine fever virus (rASFV) I/II were identified in northern Vietnam. The researchers sequenced 13 target genes from these isolates and compared them with corresponding sequences from rASFV I/II strains in China. Notably, all sequences aligned with those from China, except for the variable region (CVR), where three distinct variants were observed. This suggests that there may have been three independent introductions of rASFV I/II strains into Vietnam, potentially originating from China, as ASFV p72 genotype I strains have not been previously reported in the region.

The findings raise significant concerns regarding the efficacy of current live-attenuated ASFV vaccines licensed in Vietnam, which have been effective against the highly pathogenic ASFV p72 genotype II strains. However, the rASFV I/II strains from China appear to exhibit resistance to these vaccine candidates. Consequently, the newly identified strains in Vietnam may also be resistant to the immunity conferred by existing vaccines, posing a substantial challenge for disease management and vaccine development. This underscores the urgent need for enhanced surveillance and control measures in the global fight against African swine fever virus (ASFV).

Discussion

In this study, conducted in September and October 2023, researchers collected 26 whole blood samples from pigs suspected of African Swine Fever Virus (ASFV) infection across six northern provinces in Vietnam. Utilizing ASFV-specific real-time PCR, all samples tested positive for ASFV, with cycle threshold values ranging from 16.58 to 32.13. Subsequent analysis through conventional PCR and Sanger sequencing revealed the presence of recombinant ASFV strains, specifically genotypes I and II, in six samples. Phylogenetic analysis indicated that while the p72 sequences belonged to genotype I, the p54 sequences were classified as genotype II, and the CD2v sequences corresponded to serotype VIII.

The findings suggest that these recombinant strains may complicate ongoing ASFV control efforts in Vietnam, as they exhibit genetic characteristics similar to those found in recent ASFV outbreaks in China. Notably, the study identified three distinct CVR variants, indicating multiple independent introductions of these recombinant strains into Vietnam. Given that the current live-attenuated ASFV vaccines in Vietnam are effective against genotype II but may not confer immunity against the newly identified rASFV I/II strains, this raises significant concerns regarding disease management and vaccine efficacy in the region. The results underscore the urgent need for enhanced surveillance and adaptive strategies in the global fight against ASFV.
