Our findings indicate that L1CAM is predominantly found in larger EVs, as evidenced by its higher sedimentation in the 10,000 × g (10K) pellet compared to the 120,000 × g (120K) pellet. We employed size-exclusion chromatography (SEC) to fractionate plasma samples, revealing that the majority of L1CAM+ EVs eluted in the early SEC fractions (1-6), which were largely free from plasma proteins, particularly albumin. The presence of L1CAM was confirmed in both early and late SEC fractions, suggesting that both large and small EVs in blood carry the L1CAM ectodomain.

Additionally, we explored the potential confounding effects of soluble L1CAM in plasma, which is primarily derived from proteolytic cleavage of membrane-bound L1CAM.
