Methods
The study was conducted in the ARBOfun research arboretum near Leipzig, Germany, which spans 2.5 hectares and features 100 tree species from 39 families, planted at a spacing of 5.8 meters. Established between 2012 and 2014, the arboretum is divided into five blocks, each containing one individual of each species. The region experiences a mean annual precipitation of approximately 534.3 mm and a mean annual temperature of 9.4 °C. The soil, classified as Luvisol with a pH of 5.7, was previously utilized as a managed arable field.
For this study, 25 broadleaved species were selected based on criteria including tree diameter and the vitality of at least three individuals per species. This selection was made to facilitate the labor-intensive process of collecting and analyzing root samples from various root orders, ensuring a comprehensive examination of the chosen species’ root systems.
Results
The results indicate significant differences in root traits between absorptive and transport roots, with absorptive roots exhibiting higher specific root length (SRL) and cortex to stele ratio (C:S), while transport roots had greater root diameter (RD) and root tissue density (RTD). Correlation analyses revealed more negative relationships between RD and both RTD and leaf dry matter content (LDMC) in absorptive roots, alongside a stronger positive correlation between RD and mycorrhizal colonization rate (MCR). Principal component analysis (PCA) showed that the first two axes for absorptive root traits explained 78% of the variability, with PC1 positively associated with RD and MCR, while transport root traits explained 67% of variability, with PC1 positively associated with RD but negatively with SRL.
Linear regression analyses demonstrated that absorptive root traits were negatively correlated with tree growth (R² = 0.35, P < 0.01), indicating that trees with thinner absorptive roots and lower MCR exhibited higher growth. Conversely, transport root traits showed no significant relationship with tree growth for PC1, but PC2 indicated a significant negative correlation with C:S and MCR (R² = 0.16, P < 0.05). Leaf traits also positively correlated with tree growth (R² = 0.19, P < 0.05), with a stronger combined effect of absorptive root and leaf traits on growth (R² = 0.39, P < 0.001) compared to transport root and leaf traits (R² = 0.18, P < 0.05). Overall, absorptive root traits were found to be more influential on tree growth than transport root traits or leaf traits, with the inclusion of multiple principal components slightly enhancing predictive power.
Discussion
In this study, we investigated the functional separation of fine roots into absorptive and transport categories, revealing that trait coordination within these root types is comparable, contrary to our initial hypothesis. Key traits such as mycorrhizal colonization rate (MCR), root diameter (RD), and cortex-to-stele ratio (C:S) significantly influenced tree growth, with absorptive root traits showing a stronger correlation to growth than transport root traits. Notably, species with thinner roots and lower mycorrhizal colonization exhibited enhanced growth, suggesting that efficient soil resource exploration is crucial for tree performance. Our findings challenge the conventional view that transport roots are less colonized by mycorrhizae, as some species demonstrated significant colonization even in higher root orders.
Furthermore, the analysis indicated that absorptive root traits are more directly linked to resource acquisition, which is essential for tree growth, while transport roots primarily serve in resource transport and storage. The integration of leaf traits with root traits further elucidated the relationship between resource acquisition and tree growth, highlighting that both above- and below-ground traits are vital for optimizing growth. The study emphasizes the need for a nuanced understanding of root functional roles, advocating for future research to explore the anatomical and functional diversity of roots across different species to better predict tree growth dynamics.
