Notably, the MbTMV-MT F2 offspring exhibited a wide range of phenotypes, contrasting with the uniform traits observed in the F1 hybrids and MiMe tetraploids. This variation suggests that unreduced, clonal gametes can be produced, potentially leading to chromosome truncations due to DNA double-strand breaks that are not repaired through homologous recombination. We also explored the MiMe system for polyploid genome design, resulting in the creation of ‘4-haplotype’ (4-Hap) plants that retain the complete genetic makeup of their four inbred grandparents.

Through whole-genome sequencing, we confirmed the presence of all four parental genomes in the 4-Hap plants and validated the inheritance of mutations in MiMe target genes.
