Methods

In this study, the authors utilized Setd1a tm1a(EUCOMM)Wtsi mice, referred to as Setd1a +/-, which were backcrossed to C57BL/6 J mice for over ten generations. The colony was maintained in J. A. Gordon’s laboratory through breeding Setd1a +/- with C57BL/6 J mice. A total of 73 mice were included in the experiments, comprising both male and female Setd1a +/- (16 males, 22 females) and wildtype (WT) littermates (15 males, 20 females).

The mice were housed in a controlled environment with a 12-hour light-dark cycle and had unrestricted access to food and water, except during periods of behavioral food restriction. Prior to surgical procedures, the mice were group-housed (up to five per cage) and were subsequently single-housed following chronic electrode implantation. All experimental protocols were approved by the National Institute of Neurological Disorders and Stroke Animal Care and Use Committee, adhering to the guidelines outlined in the Guide for the Care and Use of Laboratory Animals by the National Research Council.

Contrary to initial hypotheses, Setd1a +/- mice exhibited largely preserved hippocampal-prefrontal (HPC-mPFC) synchrony, with significant abnormalities primarily in the Re-mPFC circuit. Specifically, these mice displayed reduced beta synchrony during the delay epoch and blunted modulation of both beta and gamma synchrony across task epochs. Notably, the modulation of beta-gamma synchrony from the sample to the delay epoch was predictive of successful SWM performance in WT mice, but this relationship was disrupted in Setd1a +/- mice, indicating a critical role of thalamo-prefrontal synchrony dynamics in cognitive function.

Behaviorally, Setd1a +/- mice demonstrated normal acquisition of the delayed non-match to sample (DNMS) task but showed context-dependent deficits during trials with a fixed 10-second delay, suggesting a reduced ability to adapt to stable task demands. The findings highlight that while the overall task performance may not differ significantly during variable delay conditions, the ability to optimize performance under consistent demands is impaired in Setd1a +/- mice. This study underscores the importance of thalamo-prefrontal synchrony dynamics in SWM and suggests that disruptions in these dynamics may contribute to cognitive dysfunction associated with schizophrenia. Future research is needed to explore the causal relationships between these synchrony dynamics and behavioral outcomes.
