In this study, we identify a distinct subpopulation of microglia in the aging central nervous system (CNS), termed Autophagy Dependent Age-acquired Microglia (ADAM), characterized by elevated ERK1/2 phosphorylation and enhanced autophagic activity. Through RNA expression analysis of neuroinflammation-related genes, we found that aged microglia with high ERK1/2 phosphorylation exhibited a unique transcriptional profile, including significant upregulation of autophagy-related genes such as Rala and ULK1, and markers associated with reactive microglia. Notably, the loss of ULK1 led to a marked reduction in the ADAM population, increased apoptosis, and heightened ER stress, indicating that canonical autophagy is crucial for the survival of this microglial subset.

Furthermore, we demonstrated that IL-34, a ligand for CSF1R, selectively expanded the ADAM population and conferred neuroprotective effects during autoimmune neuroinflammation, as evidenced by improved neuronal survival and reduced clinical signs in EAE models. In contrast, CSF-1 treatment did not yield similar benefits, highlighting the specific role of IL-34 in sustaining the ADAM population. The findings suggest that the ADAM microglia play a protective role in the aging CNS, and their loss exacerbates neuroinflammatory conditions, challenging the traditional view of microglial activation as solely detrimental.
