In this study, we investigated the virulence mechanisms of opportunistic Xanthomonas strains, specifically Leaf131 and Leaf148, which were found to degrade leaf tissue in Arabidopsis thaliana. Using a quantitative assay with leaf discs, we demonstrated that tissue degradation was significantly more pronounced in rbohD mutant plants compared to wild-type Col-0 plants, indicating a correlation between the absence of the NADPH oxidase RBOHD and increased susceptibility to these pathogens. The degradation process initiated at the leaf edges and progressed over time, leading to complete tissue loss and fragmentation. We also assessed the impact of various plant genotypes on tissue degradation, revealing that certain mutants lacking pattern-triggered immunity receptors exhibited susceptibility similar to rbohD plants.
Furthermore, we explored the role of the type II secretion system (T2SS) in the virulence of Xanthomonas. Our findings indicated that the T2SS Xps is essential for the secretion of cell wall-degrading enzymes, which are crucial for tissue degradation and virulence. Mutants lacking the xps operon showed significantly reduced degradation activity and virulence in planta. Additionally, we established a causal relationship between the presence of Xanthomonas Leaf131 and shifts in the microbiota composition in rbohD plants, suggesting that the virulence of Xanthomonas indirectly benefits specific commensal bacteria by promoting dysbiosis.
